How to Pour Agar Plates (Step-by-Step)

1. Cool the Agar

  1. After sterilization, remove the agar container from the pressure cooker.

  2. Allow it to cool to about 125°F (52°C)

  3. Use a temperature gun to monitor.

  4. If it cools too much, the agar will solidify before pouring.

  5. While cooling, place the container on a stir plate to keep the agar spinning.

  6. This prevents settling or clumping.


2. Prepare Your Workspace

  1. Set up in front of a flow hood or inside a still air box (SAB).

  2. Wipe down and sanitize your workspace.

  3. Remove petri dishes from their packaging so they’re ready to pour.


3. Pour the Agar Plates

  1. Keep empty dishes stacked in piles of 10—a good size for beginners.

  2. Lift the lid of the bottom plate in the stack just enough to pour.

  3. Pour agar into the dish, then replace the lid.

  4. Repeat the process for each plate until all agar is used.


4. Solidify & Store

  1. Allow plates to cool and fully solidify.

  2. Wrap dishes as preferred:

    Individually Or in stacked groups

  3. Let plates sit for 1 week to confirm they remain clean and contaminant-free before use.
